This almost seems like it is already there since it allows me to select ISO files, but I couldn't get it to work.
I make home videos, wedding videos, etc with Sony Vegas Pro. For some reason when writing for Blu Ray it outputs to ISO. HandBrake would attempt to open them and then fail. Since I just recently began to dabble in DLNA and such I will probably encode a version from it in mp4 from now on, but I still have several iso backups I'd like to give a smaller footprint on my drives.

HandBrake can open DVD ISO images, but not Blu-ray (because the library we use for BD doesn't support it at this point). So for Blu-ray images you'll have to mount it first (using third-party software), then point HandBrake to the mounted drive, rather than the ISO image file.
